Your goal is clone your favorite gene NRG1 using restriction digestion with type II restriction enzymes, followed by ligation in a pET plasmid, and transformation in E. coli competent cells.  The objective is to clone NRG1 between the NcoI and XhoI of the pET22b. This will allow NRG1 protein to be in frame with the pelB leader peptide and be transported to the E. coli periplasm (secretion pathways are cool), where NRG1 can fold properly. NRG1 can be amplified by PCR from cDNA of neuronal cells. How will you make sure that NRG1 will be ligated to the NcoI and XhoI sites of pET22b?
