I аm studying fibrоblаsts isоlаted frоm a pediatric patient who exhibited symptoms of developmental delay, microcephaly, and brittle hair and nails. I noticed that the cells showed a marked deficiency in the levels of cysteine rich proteins CRIP1 and CRIP2 as well as in the cysteine and glycine rich protein CSRP1. After sequencing the CRIP1, CRIP2, and CSRP1 genes, I did not detect any mutations in the coding sequence or regulatory region. Bulk RNA sequencing did not reveal any widespread changes in transcript levels or splicing. Using proteomic profiling I noticed decreases in the levels of other cysteine rich proteins like metallothioneins compared to control cells but did not find overall changes in total protein levels. After performing whole genome sequencing, I identified a nonsense mutation that was most likely to cause the patient’s symptoms as well as the decrease in CRIP1, CRIP2, and CSRP1 protein levels.
